Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, T helper cells, and monocytes. CD4 serves as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 recognize distinct epitopes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.1 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

NovaFluor Compatibility:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) enhances staining performance. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample.

Technology:
NovaFluor conjugates utilize Phiton technology, featuring engineered nucleic acid dye structures optimized for spectral performance and minimized spillover.

Target Information

CD4 antigen mediates recognition of MHC class II molecules and acts as a co-receptor for HIV. Expressed mainly on T-helper cells, as well as monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. CD4 participates in T-cell activation via Lck kinase association and may contribute to neuronal damage in infectious or immune-mediated CNS diseases. Multiple splice variants are known.
