Target Information

L-fucose is a key sugar in glycoproteins and other complex carbohydrates, involved in cell-cell recognition and other macromolecular functions. The fucosyl donor for these oligosaccharides is GDP-beta-L-fucose.
There are two pathways for GDP-fucose biosynthesis: the major pathway converts GDP-alpha-D-mannose to GDP-beta-L-fucose, while the alternate salvage pathway, active in tissues such as liver and kidney, reutilizes L-fucose from glycoprotein and glycolipid turnover. This pathway phosphorylates L-fucose to beta-L-fucose-1-phosphate, which then condenses with GTP by fucose-1-phosphate guanylyltransferase to form GDP-beta-L-fucose.
