Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45, also known as Leukocyte Common Antigen (LCA). CD45 is expressed by all hematopoietic cells excluding circulating erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase enzymatic activity and plays an important role in activation of lymphocytes.

Applications Tested: Pre-titrated and validated for use at 5 µL (0.25 µg) per test.
A test is defined as the antibody amount that stains a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test).

Fluorochrome Information:
Alexa Fluor® 700 emits at 723 nm and can be excited with the red laser (633 nm). Most instruments require a 685 LP mirror and 710/20 filter.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells (~10% of lymphocyte surface proteins). It is absent on non-hematopoietic cell lines and tissues.
CD45 glycoprotein regulates Src-family kinases, lymphocyte development, and antigen signaling. Isoforms arise from alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
CD45RA is one isoform with restricted expression among lymphoid subtypes. CD45 influences cell survival, integrin-mediated signaling, apoptosis, and immunological function regulation.
