Mouse anti-Pig CD163 antibody, clone 2A10/11 recognizes porcine CD163, an approximately 120 kDa single-pass type I transmembrane glycoprotein expressed on cells of the monocyte/macrophage lineage.
Target Information
CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily and acts as a receptor for the hemoglobin–haptoglobin complex. It protects tissues from oxidative damage caused by free hemoglobin and may contribute to iron recycling via endocytosis of hemoglobin/haptoglobin complexes and subsequent heme breakdown.
CD163 is expressed exclusively on monocytes and macrophages, particularly in the late phase of inflammation. It is present on all circulating monocytes and most tissue macrophages, except those in the mantle zone and germinal centers of lymphoid follicles, interdigitating reticulum cells, and Langerhans cells. CD163 expression is higher on CD16⁺ monocytes and is upregulated by IL-10, IL-6, and dexamethasone. Conversely, LPS and PMA induce shedding of CD163 into plasma or cell supernatant.
CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, showing higher affinity for multimeric haptoglobin (HP1F phenotype) than for dimeric haptoglobin (HP1S phenotype). It also triggers intracellular signaling cascades involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL6 and CSF1.
