Description:
The 1C10 monoclonal antibody reacts with mouse CD40, a 45–50 kDa type I transmembrane glycoprotein belonging to the TNFR family. CD40 is expressed on mouse B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It plays a critical role in B cell development and maturation by inducing Ig isotype switching, preventing apoptosis, and promoting proliferation. Interaction with CD154 (gp39) on T cells is essential for T-B cell communication and immune regulation.

The 1C10 antibody exhibits agonistic activity both in vitro and in vivo.

Applications Tested:
Tested in flow cytometric analysis of mouse splenocytes. Recommended ≤1 µg per test in 100 µL volume. Optimal concentration should be determined empirically (10⁵–10⁸ cells/test).

Target Information

CD40 is a single-chain glycoprotein belonging to the TNF/NGF receptor superfamily, homologous to CD30. It is expressed on all B cells except plasma cells, as well as on some epithelial cells, carcinomas, and lymphoid dendritic cells. CD40 mediates immune and inflammatory responses such as T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.

The AT-hook transcription factor AKNA regulates CD40 expression, facilitating homotypic cell interactions. TNFR2 acts as an adaptor protein mediating CD40 signal transduction. CD40-CD154 interaction is implicated in amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis. Two alternatively spliced CD40 transcripts encoding distinct isoforms have been reported. Disorders related to CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
