Description:
The HM40-3 monoclonal antibody reacts with mouse and rat CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by mouse B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells.
CD40 regulates B cell development and maturation by inducing Ig isotype switching. In combination with IL-4, it protects B cells from surface Ig-induced apoptosis and promotes proliferation. Interaction of CD40 with CD154 (gp39) on T cells is crucial for T-B cell crosstalk and immune regulation.

• Inhibition of soluble CD154 binding to CD40
• In vitro activation of B cells

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤1 µg per test (100 µL final volume). Cell count may vary (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Target Information

CD40 is a single-chain glycoprotein belonging to the TNFR/NGF superfamily and homologous to CD30. It is expressed on all B cells except plasma cells, and also on epithelial cells, carcinomas, and lymphoid dendritic cells.
CD40 plays a key role in immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
AKNA transcription factor regulates CD40 expression, enabling homotypic cell interactions. TNFR2 acts as a signal transduction mediator with CD40.
CD40-CD154 interaction is implicated in amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
