Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. The PR mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy. PR exists as a 94 kDa protein (Form A) or a 120 kDa protein (Form B) due to alternative translation initiation. PR-B is the transcriptionally active form responsible for endometrial maintenance, pregnancy, and inhibition of ovulation. PR-A lacks 165 amino acids at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity. In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon progesterone binding, conformational changes allow dimerization and binding to progesterone response elements (PRE), activating transcription. Null mutations in the PGR gene lead to reproductive abnormalities.
