Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy.
PR exists as two forms:
- Form A (94 kDa)
- Form B (120 kDa)
These forms result from alternative translation initiation sites. PR-B is transcriptionally active and regulates genes involved in endometrial maintenance, pregnancy, and ovulation inhibition. PR-A lacks 165 amino acids at the N-terminus, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, PgR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, the receptor undergoes a conformational change, dimerizes, and binds to progesterone response elements (PRE), activating transcription. Null mutations in the PGR gene lead to reproductive abnormalities.
