Product Specific Information

The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays a key role in T cell development and function. RM4-5 binding is blocked by GK1.5.

• Applications Tested: Flow cytometric analysis of mouse thymocytes and splenocytes. Recommended ≤0.125 µg/test (100 µL total volume).
• Super Bright 702 Dye: Excited by 405 nm violet laser, emits at 702 nm. Use 710/50 bandpass filter.
• Staining Panels: When using multiple Super Bright dyes, use Super Bright Staining Buffer (SB-4401-42) to minimize non-specific interactions.
• Light Sensitivity: Protect vial and samples from light.
• Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333).

Target Information

CD4 antigen is involved in MHC class II recognition and acts as a co-receptor for HIV. Expressed mainly in T helper cells, and also in monocytes, macrophages, and dendritic cells. Tissue-level expression includes thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. CD4 aids early T-cell activation via association with Lck kinase and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts identified (RefSeq, July 2017).
