Product Specific Information

Description:
The M1/69 monoclonal antibody reacts with the mouse CD24 molecule (Heat Stable Antigen, HSA). This 35–50 kDa GPI-anchored molecule is expressed on erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells. CD24 is variably glycosylated, leading to heterogeneity in molecular mass across cell types. Expression of CD24 is used to determine stages of B lymphopoiesis in mouse bone marrow. It has been reported that P-selectin (CD62P) binds to CD24.

Applications Reported:
The M1/69 antibody is suitable for flow cytometric analysis, immunoblotting (WB), and immunohistochemical staining. It has also been reported in in vitro functional assays. For functional assays, use the Functional Grade purified M1/69 (Product #16-0242-85).

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes and thymocytes. Recommended usage ≤0.125 µg per test (100 µL final volume). Optimal titration should be determined empirically (10⁵–10⁸ cells/test).

Target Information

CD24 is a two-chain glycosylphosphatidylinositol (GPI)-anchored glycoprotein expressed during multiple stages of B-cell development, from pro-B cells to mature B cells. Plasma cell expression is low or negative. CD24 is also found in most B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas. It may regulate B-cell proliferation, maturation, and autoimmunity control.
