Target Information

The transcription of many RNA polymerase II-dependent genes requires Spt3, a member of the S. cerevisiae SAGA complex.
Spt3 and Spt20 cooperate to recruit TATA-box binding protein (TBP) to core promoters, enabling TBP binding to SAGA-dependent promoters.
Null mutations in the Spt3 gene cause defects in sporulation, diploid filamentous growth, and haploid invasive growth, suggesting its role in yeast development and mating pathways.
At some promoters (e.g., yeast HO, HIS3, TRP3), Spt3 inhibits TBP binding, reducing transcription. This repression can be counteracted by GCN5, another SAGA complex member, through histone acetylation facilitating TBP/TFIIA complex formation.
