Target Information

Protein p21 is a tumor suppressor that inhibits cyclin-dependent kinases (CDK) and is transcriptionally activated by p53.
It plays a key role in the cellular response to genotoxic stress and regulates cell cycle progression by binding and inhibiting CDK1 and CDK2, blocking transition from G1 to S phase or G2 to M phase after DNA damage.
In the cytoplasm, p21 exhibits anti-apoptotic functions by binding to and inhibiting caspase 3, ASK1, and JNK.
Thus, p21 has dual roles in carcinogenesis, acting both as a tumor suppressor and an oncogene depending on the cellular context.
