This HIS.H8 monoclonal antibody reacts with the 6X polyhistidine (6X His) tag. The H-H-H-H-H-H motif is frequently fused to the N- or C-terminus of proteins to facilitate purification (e.g., nickel affinity chromatography and immunoprecipitation) and/or detection.
This antibody recognizes the 6X His tag anywhere within the protein of interest (N-terminus, C-terminus, or internal).

Applications Reported: Immunoprecipitation and western blotting.
Applications Tested: Tested by immunoblotting of reduced cell lysate expressing a His-tagged protein ≤1 µg/mL. Antibody should be titrated for optimal performance.
Purity: >90%, determined by SDS-PAGE.
Aggregation: <10%, determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

The 6x His tag is a synthetic oligopeptide of six consecutive histidine residues (HHHHHH). Commonly expressed at N- or C-terminal regions of recombinant proteins, it enables isolation or purification via immobilized metal affinity chromatography.
Epitope tagging allows detection of recombinant proteins lacking specific antibodies by fusing a known epitope. His-tagged proteins are purified using metal chelate chromatography due to histidine’s metal-binding properties. The affinity of the His-tag motif to Ni²⁺ enables efficient purification by Ni²⁺-NTA adsorbent.
His tag-specific antibodies facilitate detection or co-immunoprecipitation of tagged proteins.
