Product Specific Information

The eBio1-21 antibody reacts with human T cell leukemia/lymphoma 1 (TCL1), a 14 kDa proto-oncogene product involved in intracellular regulation of T cell signaling. TCL1 is identified as the oncogene located at the 14q32.1 chromosome breakpoint region in T-cell prolymphocytic leukemia (T-PLL), where it is overexpressed due to chromosomal rearrangements near T-cell receptor promoter/enhancer elements.

TCL1 binds to the pleckstrin homology domain of Akt (protein kinase B) family proteins, facilitating Akt dimerization and activation. This enhances serine/threonine phosphorylation of Akt substrates such as Ikk complex, mTOR, BAD, p70S6 kinase, FOXO transcription factors, and GSK3β, regulating cellular differentiation, growth, survival, and metabolism.

Besides its tumorigenic role in T-PLL, TCL1 is expressed in CD3⁻CD4⁻CD8⁻ thymic precursors, plasmacytoid dendritic cells, stimulated mature T cells, and B cells up to the germinal center stage. Aberrant TCL1 expression is associated with pre-malignant clonal T cell expansions and mature T cell tumors, and it shows regulated expression in chronic lymphocytic leukemia (CLL).

Applications Reported: intracellular staining followed by flow cytometric analysis, immunoprecipitation, Western blot, and immunohistology of paraffin-embedded sections.
Applications Tested: validated in Western blot (Daudi cell lysates) and intracellular flow cytometry (Daudi cells). Recommended ≤0.5 µg/test for flow cytometry (100 µL final volume).

Target Information

Overexpression of the TCL1 gene in humans is implicated in mature T cell leukemia, where chromosomal rearrangements juxtapose TCL1 with TCR-alpha or TCR-beta regulatory elements. In normal T cells, TCL1 is expressed in CD4⁻/CD8⁻ cells but not in later stages of differentiation. TCL1 functions as a coactivator of the cell survival kinase AKT.
