Description:
The DJR1 monoclonal antibody reacts with human DR4 (TRAIL-R1). DR4 binds to TRAIL, activates NF-kappaB, and induces TRAIL-mediated apoptosis. DR4 is expressed by various human tumor cells and at low levels on human leukocytes. Expression has also been found intracellularly in some cell lines (Zhang et al.), confirmed via intracellular staining of Jurkat cells.

Applications Tested:
Tested by flow cytometric analysis of human DR4-transfected cells, usable at ≤1 µg per test (100 µL final volume). Optimal performance requires titration based on cell number (10⁵–10⁸ cells/test).

Target Information
TRAIL-R1 (CD261, DR4) is a type I transmembrane protein, also known as TRAIL receptor 1. Its ligand, TRAIL, belongs to the TNF family. DR4 mediates apoptosis and NF-kappaB activation, similar to Fas and TNFR1 receptors. Apoptosis, a programmed cell death process, is essential for normal cellular differentiation and development. It is induced through cytokines such as TNF and Fas ligand interacting with death domain-containing receptors (TNFR1, Fas receptor).
