Target Information

Estrogen Receptor beta (ER β, NR3A2) is a ~55 kDa protein, with multiple isoforms ranging from 36–59 kDa. Initially cloned from human testis, ER β belongs to the steroid/thyroid hormone receptor superfamily.
It acts as a ligand-activated transcription factor that binds to estrogen response elements (EREs) in DNA, regulating gene transcription via coactivators or corepressors.
Two forms of estrogen receptor exist—α (ESR1) and β (ESR2)—encoded by separate genes. ER β binds estrogens with similar affinity to ER α and can inhibit ER α activity in reproductive tissues.
ER β is widely expressed and functions as a tumor suppressor, with implications in prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular disease risk.
