Target Information

KIT (c-KIT) is a proto-oncogene encoding a type 3 transmembrane receptor for mast cell growth factor (MGF), also known as stem cell factor (SCF). It was first identified as the cellular homolog of the feline sarcoma viral oncogene v-kit. KIT and its ligand regulate the growth and activation of various hematopoietic and non-hematopoietic cells.

Mutations in KIT are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism. The D816V activation loop mutation is a key therapeutic target in neoplastic mast cell growth.

c-Kit is expressed in hematopoietic stem cells, germ cells, mast cells, and gastrointestinal tract Cajal cells. Upon SCF binding, c-Kit dimerizes and undergoes autophosphorylation, enabling Grb2 docking and activation of the Ras/ERK pathway. SCF/c-Kit signaling also activates PI3K, PLC-γ, and JAK/STAT pathways, which are essential for hematopoiesis, stem cell maintenance, and gametogenesis.
