Preparation:
Produced from sera of goats immunized with highly pure Recombinant Human sRANK Ligand. Anti-Human sRANK Ligand-specific antibody was purified by affinity chromatography employing an immobilized Human sRANK Ligand matrix.

Sandwich ELISA:
To detect Human sRANK Ligand by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity-purified antibody, in conjunction with PeproTech Biotinylated Anti-Human sRANK Ligand (500-P133GBt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Human sRANK Ligand.

Western Blot:
To detect Human sRANK Ligand by Western Blot, use at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Human sRANK Ligand is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.

Target Information

This gene encodes a member of the tumor necrosis factor (TNF) cytokine family, which is a ligand for osteoprotegerin and functions as a key factor for osteoclast differentiation and activation. It acts as a dendritic cell survival factor and is involved in the regulation of T cell-dependent immune responses. T cell activation induces expression of this gene, leading to increased osteoclastogenesis and bone loss.

This protein activates antiapoptotic kinase AKT/PKB through a signaling complex involving SRC kinase and TRAF6, suggesting a role in apoptosis regulation. Targeted disruption of the related gene in mice leads to severe osteopetrosis and lack of osteoclasts, as well as defects in early differentiation of T and B lymphocytes and failure to form lobulo-alveolar mammary structures during pregnancy. Two alternatively spliced transcript variants have been identified.
