Product Specific Information
This H132 monoclonal antibody recognizes the human T cell receptor (TCR) Vβ13.2 allele. TCR specificity is determined by Valpha, Jalpha, Vbeta, Dbeta, and Jbeta gene rearrangements. Vβ expression has been studied in relation to superantigens, inflammation, autoimmune disease, and HIV infection. More recently, TCR Vβ expression has been used to phenotype T cell clonality in CD3+/TCRαβ+ large granular lymphocyte leukemias.
The H132 antibody specifically recognizes Vβ13.2 but not Vβ13.1 or Vβ13.3.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Use at 5 µL (0.125 µg) per test in a final volume of 100 µL.
Cell number may range from 10⁵ to 10⁸ cells/test.
Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
T cell receptors (TCR) discriminate foreign from self-peptides presented by MHC class II molecules, essential for adaptive immunity. Recognition of self-peptides is linked to autoimmune diseases, while mutant self-peptides are associated with tumors. Superantigen engagement of TCRs can cause toxic shock syndrome. Autoantibodies to Vβ segments have been found in rheumatoid arthritis and SLE patients, potentially modulating TH1-mediated inflammatory reactions.
Most human T cells express TCR αβ and either CD4 or CD8. A small subset lacks both (double negative, DN), increased in some autoimmune and immunodeficiency disorders. γδ T cells, found mainly in epithelium, show limited diversity and distinct antigen recognition, with subsets exhibiting antitumor and immunoregulatory activity.
