Product Specific Information

The Notch family of transmembrane receptors regulates cell-fate decisions during organ development in many species. Upon ligand binding, Notch receptors are cleaved, releasing an intracellular domain that translocates to the nucleus to suppress lineage-specific gene expression.

The mN1A antibody specifically reacts with the intracellular domain of mouse and human Notch1, but not with Notch2, 3, or 4. It shows low affinity for the full-length unprocessed Notch1 forms.

In mice, Notch1 mRNA is expressed in hematopoietic cells of the fetal liver, adult thymus, and bone marrow. Protein expression is observed in CD4-CD8- (double-negative) and CD4-CD8+ (single-positive) thymocytes. Notch1 is involved in the regulation of lymphopoiesis and myelopoiesis.

Target Information

The Notch gene family encodes transmembrane proteins with multiple cysteine-rich EGF-like repeats. Four mammalian Notch genes (Notch1–4) are involved in differentiation of the nervous system and other tissues.
Ligands Delta and Serrate interact with Notch1. Mature Notch proteins are heterodimeric receptors composed of extracellular (NEC) and transmembrane (Ntm) subunits formed by proteolytic cleavage.
