Product Specific Information
Description:
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. Together with CD3γ and CD3δ, the ε chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking via UCHT1 induces intracellular signaling leading to cellular activation and proliferation.

Applications Tested:
- Pre-titrated and validated for flow cytometry using normal human peripheral blood cells
- Recommended use: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)
Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm.
Recommended filter: 610/20 bandpass.
Use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions when using multiple Super Bright dyes.
Handling and Fixation:
- Light-sensitive: protect from light
- Fixed samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333)

Target Information
The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating TCR surface expression. It includes CD3γ, CD3δ, CD3ε, and CD3ζ, encoded on human chromosome 11. These subunits associate with intracellular signaling molecules via tyrosine-based motifs.
CD3 is present on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells.
Defects in the CD3 gene are associated with CD3 immunodeficiency.
