Product Specific Information

The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. CD3e, together with CD3γ and CD3δ, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed in thymocytes and all mature T cells. Binding of 145-2C11 to TCR initiates intracellular signaling leading to activation, proliferation, or apoptosis depending on conditions. Commonly used as a phenotypic marker for mouse T cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometry of mouse splenocytes; ≤1.0 µg/test recommended. A test is defined as staining in 100 µL volume with 10⁵–10⁸ cells. Titrate for optimal performance.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass.

Usage Notes:

• Use Super Bright Complete Staining Buffer (SB-4401) when combining multiple Super Bright dye-conjugated antibodies to reduce non-specific interactions.
• Protect vial and samples from light (photo-induced oxidation sensitive).
• Fixation: Samples can be stored up to 3 days at 4°C in dark using IC Fixation Buffer (00-8222) or Fix/Lyse Solution (00-5333).
• Excitation: 405 nm; Emission: 600 nm; Laser: Violet

Target Information

The CD3 complex transduces antigen-recognition signals in T cells and regulates TCR surface expression. It includes CD3γ, CD3δ, CD3ε, and CD3ζ subunits, encoded on chromosome 11. CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes but not on B or NK cells. CD3 defects are associated with immunodeficiency and abnormal T cell development.
