Target Information

Beta Arrestin 1 is a member of a protein family widely expressed throughout the body, particularly abundant in the central nervous system. Acting as an adaptor or scaffold molecule, it is essential for mitogenic signaling and mediates agonist-dependent desensitization and internalization of G protein-coupled receptors (GPCRs), such as the beta 2-adrenergic receptor.
Upon ligand binding and G protein interaction, GPCRs are phosphorylated by G-protein receptor kinases (GRKs) on serine residues. Cytosolic beta Arrestin 1 is phosphorylated by ERK1/2 on serine 412 in a negative feedback mechanism and binds to phosphorylated receptors at the plasma membrane. Subsequent dephosphorylation of serine 412 leads to GPCR internalization and activation of the Ras–Raf–ERK1/2 signaling pathway.
