Product Specific Information

  • This antibody does not react with Mouse or Rat.
  • For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation.
p53 up-regulates stress response genes such as p21waf1, contributing to G1 or G2/M arrest. Structurally, it contains an N-terminal transactivation domain, a central DNA-binding domain, an oligomerisation domain, and a C-terminal regulatory domain.
Phosphorylation at Ser15 is critical for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation.
p53 is expressed at low levels in normal cells but elevated in transformed cell lines, contributing to malignancy. Mutations in TP53 disrupt DNA binding and tumor suppressor activity, commonly found in human cancers and in germline mutations such as Li-Fraumeni syndrome.
