Description:
SPRCL5 monoclonal antibody reacts with Mouse CD185 (CXCR5), also known as C-X-C chemokine receptor 5 or Burkitt lymphoma receptor 1 (BLR1). CXCR5 is a seven-transmembrane G protein-coupled receptor originally identified in Burkitt’s lymphoma. In peripheral blood, CXCR5 is expressed on B cells, CD4+ T cells (not Th1 or Th2), and a subset of memory (CD45RO+) T cells.

CXCR5+ circulating T cells are in a resting state and migrate to lymph nodes due to CCR7 and CD62L expression. In tonsil, CXCR5 is expressed on nearly all CD4+ cells along with CD45RO and activation markers such as CD69 and ICOS. These cells can induce antibody production when co-cultured with B cells, supporting their role in B cell help. CXCR5 plays a crucial role in lymphocyte trafficking, particularly T cell migration into B cell follicles in response to CXCL13, marking follicular helper T cells (Tfh).

Note: SPRCL5 does not recognize formaldehyde-fixed epitopes; staining should be performed prior to fixation.

Applications Tested: Tested on mouse splenocytes. Use ≤0.25 µg/test (in 100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.

CXCR5 (C-X-C Chemokine Receptor Type 5, CD185), also known as Burkitt lymphoma receptor 1 (BLR1), belongs to the CXC chemokine receptor family. It is a G protein-coupled seven-transmembrane receptor for chemokine BLC, expressed in lymphatic tissues such as lymph node follicles and spleen. CXCR5 plays an essential role in B cell migration and is expressed in various tissues including blood, bone marrow, brain, liver, lymph node, skin, spleen, stomach, and tonsil.

G-protein Coupled Receptors (GPCRs) share a structural motif of seven membrane-spanning helices and exist in active/inactive forms. Ligand binding triggers signaling via G-proteins and downstream effectors. GPCR regulation mechanisms are key research areas in pharmacology, with several GPCR-targeted drugs developed for mental health and allergy treatments.
