Product Specific Information

The monoclonal antibody HE12 recognizes human Cyclin E. Cyclins regulate the cell cycle by binding and activating cyclin-dependent kinases (CDK). Cyclin E regulates the transition from G1 to S phase by binding and activating CDK2. The Cyclin E/CDK2 complex phosphorylates downstream proteins facilitating the G1 to S phase transition. Overexpression of Cyclin E is found in several cancer types including breast, colon, bladder, lung, and skin. In breast cancer, Cyclin E overexpression correlates with increased stage, grade, and poor prognosis.

Applications Reported:
This HE12 antibody is reported for use in immunohistochemical staining of FFPE tissue sections and microscopy.

Applications Tested:
Tested by immunohistochemistry of FFPE human tissue using low pH antigen retrieval at ≤10 µg/mL. Antibody titration is recommended for optimal performance.

Fluorophore Information:
eFluor® 660 replaces Alexa Fluor® 647, emits at 659 nm, and is excited with a red laser (633 nm). Ensure instrument compatibility for detecting this fluorochrome.
Excitation: 633–647 nm; Emission: 668 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

Cyclin E is a member of the conserved cyclin family, regulating CDK kinases and cell cycle progression. It forms a complex with CDK2, essential for G1/S transition. Cyclin E accumulates at the G1–S boundary and is degraded during S phase. Overexpression is linked to tumorigenesis and chromosomal instability. It associates with NPAT protein to regulate histone gene expression and cell-cycle progression. Multiple splice variants exist.
