Product Specific Information

This me1B3 monoclonal antibody reacts with human epidermal growth factor receptor (EGFR), also known as ErbB1 and Her1. EGFR is a receptor tyrosine kinase belonging to the EGFR/ErbB receptor subfamily, which also includes ErbB2/Her2/neu. EGFR contains an extracellular ligand-binding domain, a single transmembrane region, and a cytoplasmic tail with a tyrosine kinase domain.

Specific ligands include EGF and TNF alpha, as well as other members of the EGF protein family. Upon ligand binding, EGFR undergoes receptor-mediated dimerization with itself or ErbB2/Her2/neu, leading to auto-phosphorylation and activation of PLCγ, JAK/STAT, MAPK/ERK, and PI3K/AKT signaling pathways. EGFR is involved in cell proliferation, migration, and differentiation, and is evolutionarily conserved. It is broadly expressed and overexpressed or mutated in glioblastomas and various epithelial cancers, including non-small cell lung and colorectal cancers.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested on HeLa cells. Use at 5 µL (0.125 µg) per test in a final volume of 100 µL. Cell number may vary from 10⁵ to 10⁸ cells/test.

Target Information

EGFR (epidermal growth factor receptor) is a receptor tyrosine kinase that signals in response to various growth factors. Overexpression has been linked to numerous cancers, and EGFR is the target of biological and small-molecule therapeutics. The EGFR gene is located on chromosome 7 in humans and belongs to the HER/ErbB family (ErbB1–4). Ligand binding (EGF, TGFα) induces dimerization and autophosphorylation, activating downstream signaling. Overexpression is observed in tumors of the head and neck, brain, bladder, stomach, breast, lung, endometrium, cervix, vulva, ovary, esophagus, and squamous cell carcinoma.
