Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, regulating cell death and stress responses. TRAF proteins share a homology region enabling receptor and TRAF interactions, activating various signaling cascades.
TRAF2 and TRAF3 bind directly to CD40, crucial for NF-κB activation in B lymphocytes. TRAF2, together with TRAF6, participates in CD40 signaling in non-hematopoietic cells. Additionally, TRAF2 interacts with lymphotoxin-beta receptor (LTβR) alongside TRAF3 and apoptosis inhibitors cIAP1 and Smac.
