Target Information

Varicella Zoster Virus (VZV), a member of the human herpesvirus family, causes two distinct clinical manifestations: childhood chickenpox (Varicella) and shingles (zoster). Varicella results from primary infection with VZV, whereas zoster arises from reactivation of latent virus in sensory ganglia, typically in aging or immunosuppressed individuals.

Glycoprotein E (gE), a 623-amino-acid type I integral membrane protein, is expressed on plasma membranes and in the cytoplasm of VZV-infected cells, likely residing in intracellular vesicle or organelle membranes. VZV gE complexes with gI in the rough endoplasmic reticulum, forming gE–gI heterodimers. gE is a predominant component of the virion envelope and is essential for VZV replication (encoded by ORF68). In contrast, gE genes can be deleted from herpes simplex and pseudorabies viruses, though with reduced infectivity. Since VZV lacks a gD homologue, gE or the gE–gI complex may perform functions typically divided between gD and gE in other alphaherpesviruses.
