Product Specific Information

The M17/4 monoclonal antibody reacts with mouse CD11a, the 180 kDa integrin alpha L, also known as lymphocyte function-associated antigen-1 (LFA-1) alpha chain. LFA-1, formed by non-covalent association of CD11a with CD18 (integrin beta 2), acts as an adhesion molecule involved in lymphocyte and granulocyte function. CD11a is expressed by all leukocytes and interacts with ligands CD54 (ICAM-1), CD102 (ICAM-2), and CD50 (ICAM-3).

Applications Reported:
The M17/4 antibody has been reported for use in flow cytometric analysis, immunoprecipitation, and immunohistochemical staining. It has also been used for blocking CD11a function (functional assays should use Functional Grade purified M17/4, Product #16-0111).

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes, recommended ≤0.5 µg per test (100 µL final volume). Optimal titration is recommended for best assay performance.

Target Information

CD11a (LFA-1 alpha, ITGAL) together with CD18 forms LFA-1 (alphaLbeta2 integrin), crucial for leukocyte adhesion and costimulatory signaling. CD11a interacts with ICAMs 1–3 and transitions between low- and high-affinity conformations upon cell activation. It plays roles in immune cell migration, adhesion, differentiation, and T-cell effector function. Blocking CD11a is a therapeutic strategy in inflammatory diseases; e.g., Efalizumab (Raptiva) targets CD11a to modulate T-cell migration. Simvastatin also binds CD11a, influencing immune modulation. Two transcript variants exist; dysfunctions are linked to benign cephalic histiocytosis and leukocyte adhesion deficiency.
