Product Specific Information

The eBioKMC8 monoclonal antibody reacts with mouse CD9, a 24 kDa member of the transmembrane 4 superfamily characterized by four hydrophobic membrane-spanning domains and short cytoplasmic tails. CD9 is expressed by multiple cell types including monocytes, macrophages, platelets, B and T cells, dendritic cells, eosinophils, basophils, endothelial cells, myoblasts, and neuroblasts. On T cells, CD9 acts as a co-stimulatory molecule on naïve T cells. CD9 expression is also found in oocytes, where deficiency causes sterility due to defective gamete fusion.

In mouse macrophages, CD9 associates with Fcγ receptors to modulate phagocytosis and inflammatory responses. In B cells, CD9 serves as a marker for marginal zone B cells, B1 cells, and plasma cells. In dendritic cells, it facilitates the association of heterologous MHC II molecules. Expression levels may vary among donors.

Applications Reported: Flow cytometry and Western blot (non-reducing).
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes and bone marrow cells. Recommended ≤1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.
Purity: >90% (SDS-PAGE).
Aggregation: <10% (HPLC).
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD9 antigen is a tetraspanin glycoprotein expressed on developing B lymphocytes, platelets, monocytes, eosinophils, basophils, stimulated T lymphocytes, neurons, and glial cells. It participates in cell activation and aggregation, often through integrin CD41/CD61 interactions. CD9 is involved in processes such as cell motility, osteoclastogenesis, neurite outgrowth, myotube formation, and sperm-egg fusion. It plays roles in adhesion, proliferation, and MHC II association on dendritic cells, crucial for T cell stimulation. CD9 expression contributes to suppression of cancer cell motility and metastasis, and is considered a metastasis suppressor in solid tumors.
