Product Specific Information

This antibody binds the FG loop of human CD47, which interacts with SIRP alpha and other known CD47/SIRP alpha-blocking monoclonal antibodies. CD47 is broadly distributed on normal adult tissues and ovarian tumors, and is abundant in certain epithelia and brain regions. Integrin-associated protein (IAP) acts as an adhesion receptor for THBS1 on platelets and modulates integrins. It plays a role in memory formation and synaptic plasticity in the hippocampus, and acts as a receptor for SIRPA, preventing maturation of immature dendritic cells and inhibiting cytokine production by mature dendritic cells.

Target Information

CD47 (integrin-associated protein, IAP) is a ubiquitously expressed transmembrane glycoprotein that interacts with several integrins and regulates their functions. Engagement of CD47 modulates signaling pathways, including activation of heterotrimeric G proteins and increases intracellular calcium concentration upon cell adhesion to extracellular matrix. CD47 serves as a receptor for the C-terminal cell binding domain of thrombospondin and may play a role in membrane transport and signal transduction. It is broadly distributed across tissues but reduced on Rh erythrocytes. CD47 acts as a ligand for CD172a (SIRP alpha), an immune inhibitory receptor on macrophages, preventing phagocytosis of CD47-positive cells. The CD47-CD172a system influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanosensing, and neuronal development, being abundant in synapse-rich brain and retinal regions.
