Product Specific Information

The II/41 monoclonal antibody reacts with the mu heavy chain of mouse IgM. It does not react with other mouse immunoglobulin classes (IgD, IgG, IgA).
IgM is expressed intracellularly during early B cell development and on the surface of mature B cells. Fluorochrome-conjugated II/41 can be used as a detection secondary for mouse IgM.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse bone marrow cells, ≤ 1.0 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Antibody should be titrated for optimal performance.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor applications.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample (regardless of number of NovaFluors).
For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures designed to minimize spectral spillover and spread.
Excitation: 552 nm; Emission: 612 nm; Laser: 561 nm (Yellow).

Strain-specific Note:
NovaFluor Yellow 610 shows non-specific B cell staining in SJL mice. No non-specific staining observed in BALB/c or C57BL/6 mice. Other strains not tested.

Target Information

Mouse IgM is the first antibody produced during an immune response and is the largest immunoglobulin in serum.
Composed of five heavy and five light chains, it mediates complement activation and antigen agglutination.
Crucial for innate immunity and pathogen neutralization.
Commonly used in research applications such as ELISA, Western blotting, and immunohistochemistry.
