Product Specific Information

The II/41 monoclonal antibody reacts with the mu heavy chain of mouse IgM. It does not react with other classes of mouse immunoglobulin including IgD, IgG, or IgA. IgM is expressed intracellularly during early stages of B lymphopoiesis and on the surface of mature B cells in bone marrow and peripheral tissues.

Fluorochrome-conjugated II/41 can be used as a detection secondary for mouse IgM.
Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse bone marrow cells. Recommended ≤1.0 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

NovaFluor conjugates are shipped with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01). Add 5 µL per sample to antibody cocktails/master mixes before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Internal testing shows NovaFluor Yellow 610 may non-specifically stain B cells in SJL mice; no such staining observed in BALB/c or C57BL/6 strains.

Target Information

Mouse IgM is the first antibody produced during an immune response and the largest immunoglobulin in serum. It consists of five heavy and five light chains, playing a key role in complement activation and antigen agglutination. Widely used in research applications such as ELISA, Western blotting, and immunohistochemistry.
