Description:
Monoclonal antibody 2D3 recognizes CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein with a small alternatively spliced cytoplasmic domain. It interacts with SIRP (signal regulatory protein) and integrins (CD51/CD61, CD41/CD61), mediating bi-directional signaling that regulates neural synaptic activity and macrophage phagocytosis. CD47 is a receptor for thrombospondin and is expressed in most hematopoietic and non-hematopoietic cells.

Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for use in normal human peripheral blood cells. Use 5 µL (0.6 µg) per test (100 µL total volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD47 (integrin-associated protein, IAP) is a transmembrane glycoprotein that interacts with integrins and regulates multiple signaling pathways, including G protein activation and calcium flux upon cell adhesion. It serves as a receptor for thrombospondin and modulates cell migration, adhesion, and immune inhibition by binding to CD172a (SIRPα) on macrophages, preventing phagocytosis of CD47-positive cells. CD47 also influences T and B cell activation, neuronal development, and chondrocyte responses to mechanical stimuli.
