Target Information

Three distinct signaling pathways lead to programmed cell death (apoptosis). The death receptor and mitochondrion pathways are the main ones, involving key apoptotic proteases caspase-8 and caspase-9, respectively.
The endoplasmic reticulum (ER) stress pathway is the third apoptotic mechanism, in which caspase-12 plays a role. Caspase-12 is localized to the ER but not to the cytoplasm or mitochondria. It is activated by ER stress, including disruption of ER calcium homeostasis, and mediates ER stress-induced apoptosis.
Caspase-12 co-localizes with ER proteins associated with Alzheimer’s disease, such as γ-secretase presenilin and β-amyloid precursor protein (APP), and mediates cytotoxicity induced by amyloid-β. Caspase-12 is ubiquitously expressed in mouse tissues.
