OMA1-06020 detects smoothelin from human, canine, feline, porcine, chicken, and monkey tissues. This antibody recognizes both visceral and vascular smoothelin isoforms and does not cross-react with rat samples.
It has been successfully used in Western blot, immunocytochemistry, and immunohistochemistry procedures.
By Western blot, this antibody detects ~59 kDa (visceral smoothelin) and ~100 kDa (vascular smoothelin) proteins from canine smooth muscle extracts.
Useful for discriminating fully differentiated (contractile) smooth muscle cells from proliferative ones.
For formalin-fixed paraffin-embedded tissues, boiling in 10 mM citrate buffer (pH 6.0) for 3×5 min prior to immunostaining is recommended.
Antigen: cytoskeletal smoothelin extract of chicken gizzard.
Store at 4ºC, or in small aliquots at -20ºC.

Target Information

Smoothelin is a constituent of the smooth muscle cell (SMC) cytoskeleton and is exclusively expressed in fully differentiated (contractile) SMCs.
Antibodies against smoothelin are useful tools to monitor SMC differentiation.
RNA and protein analyses show broad species distribution.
Cells with SMC-like characteristics (e.g., myofibroblasts, myoepithelial, skeletal, and cardiac muscle cells) do not contain smoothelin.
Confocal microscopy and transfection studies demonstrate a filamentous organization distinct from desmin and vimentin, with smoothelin colocalizing with actin stress fibers.
Two isoforms have been identified:

• 59 kDa (visceral SMC)
• 100 kDa (vascular SMC)
  Human smoothelin is encoded by a single-copy gene located on chromosome 22.
