Target Information
Estrogen Receptor beta (ERβ, NR3A2) is a nuclear receptor protein (~55 kDa, isoforms 36–59 kDa) initially identified in human testis. ERβ functions as a ligand-activated transcription factor that binds estrogen, inducing dimerization and DNA binding to estrogen response elements (EREs). This regulates gene transcription via coactivator or corepressor recruitment. Two forms exist—ERα (ESR1) and ERβ (ESR2)—with ERβ showing similar estrogen affinity but potentially inhibitory effects on ERα activity in reproductive tissues. ERβ is broadly expressed and acts as a tumor suppressor, relevant to prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular disease risk.
