Product Specific Information

The tbykt monoclonal antibody reacts with the human Estrogen Receptor alpha (ERα) molecule.
Estrogen receptor belongs to the nuclear receptor superfamily, functioning as a ligand-dependent transcription factor. Upon estrogen binding, ERα dissociates from Hsp90, dimerizes (with ERα or ERβ), and binds DNA directly through estrogen response elements (EREs) or indirectly via transcription factors such as Sp1, AP-1, NF-κB, and FoxA1.
ERα binding to DNA can either upregulate or repress gene expression. The presence of ERα is associated with improved survival and better response to endocrine therapy (e.g., tamoxifen) in breast cancer.

Applications Reported:

  • Western blotting
  • Immunohistochemical staining (formalin-fixed paraffin-embedded tissue)
  • Immunocytochemistry

Applications Tested:

  • IHC (P): ≤0.5 µg/mL (low/high pH antigen retrieval)
  • ICC/IF: ≤1.0 µg/mL (fixed and permeabilized human cells)
  • WB: ≤5.0 µg/mL (reduced HeLa cell lysate)

It is recommended to titrate the antibody for optimal performance in the assay of interest.

Target Information

Estrogen Receptors (ER) are nuclear receptors belonging to the steroid/thyroid hormone receptor superfamily.
When bound to estrogen, ER undergoes conformational change, dimerizes, and binds to estrogen response elements (EREs) in DNA, regulating gene transcription through coactivator or corepressor recruitment.

  • Two main forms: ERα (ESR1) and ERβ (ESR2)
  • Multiple ERα isoforms: 66 kDa, 53 kDa, 47 kDa, 35 kDa
  • Widely expressed in various tissues, essential for sexual development and reproductive function
  • Involved in pathological processes such as breast cancer, endometrial cancer, and osteoporosis
