Product Specific Information

Monoclonal antibody HFR1 recognizes human and mouse epidermal growth factor receptor 4 (ErbB4 or HER4). ErbB4 is a receptor tyrosine kinase (RTK) involved in proliferation, differentiation, migration, survival, and apoptosis, and is often dysregulated in cancer.
ErbB4 contains extracellular ligand-binding domains, a transmembrane region, and a cytoplasmic tail with tyrosine kinase domain and the HFR1 binding site. Upon ligand binding, ErbB4 dimerizes, becomes phosphorylated, and can be cleaved for nuclear translocation.
HFR1 recognizes membrane-bound, cytoplasmic, and nuclear ErbB4. Cytosolic expression correlates with positive prognosis in breast cancer, while nuclear expression inversely correlates with tumor grade and aggressiveness.

Applications Reported: Immunoprecipitation, Western blotting, IHC (formalin-fixed paraffin-embedded tissue), and ICC.
Applications Tested: ICC on A431 cells and IHC on FFPE tissue (≤10 µg/mL). Optimize dilution for best performance.

Target Information

ERBB4 (HER4) is a type I receptor tyrosine kinase related to EGFR, ERBB2, and ERBB3. It binds neuregulins and NTAK, mediating mitogenesis and differentiation. ErbB4 regulates cellular metabolism, transcription, cell cycle, cytoskeletal rearrangement, movement, apoptosis, and differentiation.
Highly expressed in breast carcinoma cell lines (T47-D, MDA-MB-453, BT-474, H3396) and normal tissues (skeletal muscle, heart, pituitary, brain, cerebellum). Expression is low or absent in MDA-MB-231, MDA-MB-157, MDA-MB-468, and SKBR-3. Elevated ErbB4 levels may contribute to tumorigenesis.
