Target Information

Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for programmed cell death protein 1 (PD-1). PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Its expression is upregulated by cytokines such as IL-4, TNF-alpha, and IFN-gamma. PD-L2 has a higher affinity for PD-1 compared to PD-L1 and functions as an inhibitory immune checkpoint molecule, regulating T cell activation, proliferation, and cytokine secretion. Additionally, PD-L2 can interact with another receptor, repulsive guidance molecule b (RGMb), influencing respiratory tolerance via BMP and neogenin signaling pathways. The human PD-L2 monomer comprises a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
