Target Information

Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. PD-L2 acts as a ligand for PD-1, along with PD-L1, and is primarily expressed in dendritic cells. Its expression can be induced in various immune and non-immune cells depending on the microenvironment, particularly by cytokines such as IL-4, TNF-alpha, and IFN-gamma.
PD-L2 has a 2–6 times higher affinity for PD-1 compared to PD-L1. The PD-1/PD-L1/PD-L2 pathway serves as an inhibitory immune checkpoint, regulating T-cell activation and cytokine secretion. Additionally, PD-L2 can stimulate T-cell proliferation even in PD-1-deficient cells, suggesting additional receptors such as repulsive guidance molecule b (RGMb).
The interaction between PD-L2 and RGMb contributes to respiratory tolerance regulation via BMP and neogenin signaling pathways. The human PD-L2 monomer consists of a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
