Target Information

Caspase-activated DNase (CAD) cleaves chromosomal DNA during apoptosis. Apoptosis, a programmed cell death process that removes toxic or unnecessary cells, can be induced by various stimuli such as tumor necrosis factors, UV irradiation, anticancer drugs, and deprivation of survival factors.
During apoptosis, cells and nuclei shrink and fragment, and chromosomal DNA is degraded into nucleosomal units. CAD, activated by caspases, exists as a complex with its inhibitor ICAD. ICAD assists in the proper folding of CAD and inhibits its DNase activity.
When caspases are activated, they cleave ICAD, releasing CAD to enter the nucleus and degrade chromosomal DNA. The activation of caspases is regulated by the balance between pro- and anti-apoptotic proteins of the Bcl-2 family, heat shock proteins, and inhibitors of apoptosis proteins (IAPs).
Currently, fourteen members of the caspase family have been identified, seven of which mediate apoptosis.
