Product Specific Information
The monoclonal antibody 29A1.4 recognizes mouse NKp46 (CD335), a member of the natural cytotoxicity receptor (NCR) family. CD335 is a glycoprotein with two Ig-like domains and a short cytoplasmic tail, expressed specifically on NK cells (including immature NK cells, DX5– CD3–). This allows discrimination between NKT cells and NK cells (NKp46+, CD3–).
Staining with 29A1.4 is not strain-specific and has been validated on C57Bl/6, SJL, CBA/CA, and BALB/C strains. NKp46 plays a role in NK cell-mediated lysis of tumor and pathogen-infected cells.
- The 29A1.4 antibody activates NK cells in vitro but does not deplete NK cells in vivo.
- Reported application: Flow cytometric analysis.
- Tested application: Flow cytometric analysis of mouse splenocytes (≤1 µg per test, 100 µL final volume).
- Recommended to titrate for optimal assay performance.
eFluor® 660 replaces Alexa Fluor® 647, emits at 659 nm, and is excited with the red laser (633 nm).

Target Information
NKp46 (CD335, NCR1) is a cytotoxicity-activating receptor that enhances the efficiency of activated NK cells in mediating tumor cell lysis. It consists of two Ig-like domains related to leukocyte immunoglobulin-like (LIR) and killer inhibitory receptors (KIR).
