Description:
The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), a low-affinity receptor for IgG with an apparent molecular weight of 50–80 kDa.
CD16 includes two similar genes:

• CD16A (FcγRIIIA): Expressed in macrophages and NK cells (polypeptide-anchored form)
• CD16B (FcγRIIIB): Expressed in neutrophils (GPI-anchored form)

There are two known polymorphisms of CD16B (NA-1, NA-2). Individuals homozygous for NA-2 show lower phagocytic capacity compared with NA-1. CD16 binds IgG immune complexes, showing preferential binding to IgG1 and IgG3, and minimal binding to IgG2 and IgG4. Upon binding, CD16 isoforms initiate signal transduction cascades leading to ADCC, phagocytosis, degranulation, and proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Tested:

• Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells
• Recommended use: 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample prior to combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).
