Peroxisomes are subcellular organelles essential for lipid metabolism, present in nearly all eukaryotic cells. The peroxisomal membrane contains integral and peripheral membrane proteins responsible for importing matrix proteins and transporting metabolites.

Pxmp2 (Peroxisomal membrane protein 2), also known as PMP22, is a 195-amino-acid multi-pass membrane protein belonging to the PXMP2/4 family. It interacts with Peroxin 19 and Siva proteins. Peroxin 19 is required for peroxisome membrane synthesis and binds multiple peroxisomal membrane proteins (PMPs).

Pxmp2 is the most abundant peroxisomal membrane protein in higher eukaryotes, with tissue-specific expression highest in liver, kidney, and heart. It is suggested to have pore-forming activity contributing to peroxisomal membrane permeability. Disruption of the Pxmp2 gene in mice restricts peroxisomal membrane permeability to solutes. The Pxmp2 gene is conserved across multiple species and maps to human chromosome 12q24.33.
