Product Specific Information
The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40, a member of the TNFR family, is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype-switching and, together with IL-4, protects B cells from apoptosis and promotes proliferation.
Interaction of CD40 with CD154 (gp39), its ligand on T cells, is crucial for T–B cell crosstalk and immune regulation. The 5C3 clone is reported to activate B cells in in vitro functional assays.

Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.06 µg) per test.
A test is defined as the amount of antibody that stains a cell sample in a 100 µL final volume.
Cell number may range from 10⁵ to 10⁸ cells/test.

Target Information
CD40 is a single-chain glycoprotein belonging to the TNF/NGF superfamily and shows homology to CD30. It is expressed on all B cells except plasma cells, as well as on some epithelial and dendritic cells. CD40 mediates immune and inflammatory responses including T cell-dependent Ig class switching, memory B cell development, and germinal center formation.
AT-hook transcription factor AKNA regulates CD40 expression, while TNFR2 serves as a signal transduction mediator. CD40–CD154 interaction is implicated in amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis. Two alternatively spliced CD40 transcript variants are known.
