Product Specific Information
For reconstitution, add 100 µL distilled water to achieve a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step using Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL (Product #89882).
Target Information
Programmed death-ligand 2 (PD-L2), also known as B7-DC, belongs to the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for programmed cell death protein 1 (PD-1).
PD-L2 is mainly expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Expression is upregulated by IL-4, TNF-α, and IFN-γ.
Although expressed at lower levels than PD-L1, PD-L2 has 2–6 times higher affinity for PD-1. The PD-1/PD-L1/PD-L2 axis serves as an inhibitory immune checkpoint, reducing T cell activation and cytokine secretion.
PD-L2 can also stimulate T cell proliferation independently of PD-1, suggesting additional receptors such as repulsive guidance molecule b (RGMb). PD-L2–RGMb interaction plays a role in respiratory tolerance via BMP and neogenin signaling pathways.
The human PD-L2 monomer comprises a 201-amino-acid extracellular domain, 21-amino-acid transmembrane domain, and 32-amino-acid cytoplasmic domain.
