For reconstitution, we recommend adding 100 µL distilled water to reach a final antibody concentration of about 1 mg/mL.
To use this carrier-free antibody for conjugation experiments, perform another round of desalting using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).
Target Information
Programmed death-ligand 2 (PD-L2, also known as B7-DC) is a member of the B7 ligand family within the immunoglobulin superfamily. It acts as a ligand for programmed cell death protein 1 (PD-1), similar to PD-L1.
PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Its expression is upregulated by Th2 cytokine IL-4 and to a lesser degree by Th1 cytokines TNF-α and IFN-γ.
Although PD-L2 is generally expressed at lower levels than PD-L1, it has 2–6 times higher affinity for PD-1. The PD-1/PD-L1/PD-L2 axis functions as an inhibitory immune checkpoint, reducing T-cell activation and cytokine secretion (e.g., IFN-γ, IL-10).
Recent studies show PD-L2 also interacts with a second receptor, repulsive guidance molecule b (RGMb), involved in respiratory tolerance through BMP/neogenin signaling pathways.
Human PD-L2 consists of a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
