Target Information
Spleen tyrosine kinase (SYK), along with ZAP70, are cytoplasmic tyrosine kinases that are potential drug targets for allergic and autoimmune diseases. SYK may also play a role in cancer.
Upon B cell antigen receptor (BCR) or high-affinity IgE receptor (FcεRI) activation, Syk translocates to the plasma membrane and phosphorylates downstream adaptor proteins. This provides docking sites for signaling pathways involved in calcium mobilization, cytoskeleton remodeling, and gene transcription.
Syk binds receptor assemblies via its SH2 domains to phosphorylated ITAM motifs, which are modified by Src-family kinases that also activate Syk through phosphorylation of its activation loop.
